Charles Piutau will play at centre for the Blues against the Cheetahs in Bloemfontein on Saturday, with Francis Saili a casualty of his team's loss to the Stormers last weekend.
Saili has been dropped to the bench, with Piutau, who plays wing or fullback for the All Blacks, moving to centre alongside Hamish Northcott, who will wear the No12 jersey.
Melani Nanai, a wider training group player, will make his debut on the right wing, with Tevita Li on the other and Lolagi Visinia named at fullback.
The re-shuffle by John Kirwan will be an attempt to find an attacking spark which has been missing in their opening defeats to the Stormers and Chiefs.
Steven Luatua has recovered from the shoulder injury which forced him from the Newlands field in the first half of the 27-16 defeat, but will start from the reserves bench. Luke Braid will wear the No6 jersey, alongside skipper and No8 Jerome Kaino, who will be playing his 100th game for the Blues, and openside Brendon O'Connor.
Inside back Dan Bowden has returned to New Zealand after aggravating a foot injury.
