Dolly labelled the other woman in the photo "Jolene", the boyfriend "my man" and the woman "me" to echo the lyrics of the country song about flirting and infidelity.
The 1973 karaoke favourite was penned by Parton after a flame-haired temptress made eyes at her husband Carl Dean shortly after the pair were wed.
And the name Jolene came from a young girl who asked the 9 to 5 singer for an autograph at a concert.
For social media users the idea that Dolly Parton is engaging with memes was almost too much to take.
One fan wrote: "Dolly Parton is tweeting memes. I repeat, the Queen is tweeting memes. What a time to be alive."
And another added: "And just like that, Dolly Parton now owns Twitter."
The original stock photo, taken in Girona in 2015, was snapped by Antonio Guillem, 45, and carried the caption "Disloyal Man Walking With His Girlfriend and Looking Amazed at Another Seductive Girl".
Guillem admitted that he had no clue the image had made such a stir. He said: "The models discovered the meme on social media and they told me about it. None of us could imagine the media repercussion that it has reached right now."
