"I said: 'What about my resume? They said: 'We don't go off of resumes," the actress told fans in her video.
She then asked what made Schumer worth millions and Netflix allegedly cited the star's two sold-out shows at Madison Square Garden and a hit summer movie.
"Is that not Amy Schumer's resume?" she said.
According to Mo'Nique, Netflix told her they believe she's a "legend" in her field.
"Then why shouldn't I get what the legends are getting?" she wondered. "Please stand with me in this boycott of Netflix."
The streaming service, which is currently airing a Mo'Nique special called I Coulda Been Your Cellmate, has not addressed the star's claims.
This isn't the first time Mo'Nique has commented on pay disparity. Last year, she lashed out at Oprah Winfrey, Tyler Perry and Lee Daniels for not paying her fairly.
"I know they like to say: 'Mo'Nique, you talk too motherf***ing much'. It would kill me not to say the real s***. You are not paying me equally. You are not treating me fairly, so you can suck my d*** if I had one," she said during a May show at the Apollo Theater.
