The Mad Men actor will go under the knife to remove a single polyp - a growth of abnormal tissue - his representative confirms to E! News.
The 42-year-old star was overheard talking about the procedure with his on-screen ex-wife January Jones at an Emmy Awards after party on Sunday night.
Jon - who is in a long-term relationship with his Friends with Kids co-star and director Jennifer Westfeldt - reportedly became alarmed after coughing up blood recently, according to Life & Style magazine.
The 42-year-old actor is said to have sought treatment immediately and discovered he has a polyp growth on his vocal chords.
Meanwhile, Jon and the rest of the Mad Men cast left this year's Emmy Awards empty handed after several years of dominating the awards and Jon recently revealed how he and Parks and Recreation comedienne Amy Poehler have decided to throw a loser's after party.
He said: "We figured that the winners get celebrated enough, so we thought it was about time that the losers get celebrated."
